Modeling the Budding Yeast Cell Cycle

Predictions (xi):

net1-ts can be arrested in nocodazole if Clbs are over-expressed but not if Clns are over-expressed or CKIs are deleted.

Genotype Phenotype
In nocodazole:

net1-ts

net1-ts 2X CLB2 (or 2X CLB5)

net1-ts 2X CLN2 (or 2X CLN3)

net1-ts sic1∆ (or cdc6∆2-49)

Loses viability (Visintin et al., 1999)

Retains viability (prediction).

Loses viability (prediction).

Loses viability (prediction).

net1-ts cannot be arrested in nocodazole. It exits from mitosis, resulting in losing viability when the drug is removed.

  • net1-ts with an extra copy of genomic CLB2 (or CLB5) can be arrested in nocodazole. Clb5 and Clb2 levels are very important in the battle against Cdc14 for Cdh1 activation. When adding an extra copy of the genes, the mutants retain viability.

  • net1-ts in nocodazole loses viability even with an extra copy of genomic CLN2 (or CLN3).

  • net1-ts in nocodazole loses viability even with additional sic1∆ (or cdc6∆2-49 or both)
